Physiological responses and transcriptome analyses of upland rice following exposure to arsenite and arsenate

This study examines the molecular response of upland rice when exposed to different inorganic arsenic (iAs) species. One-week-old (Oryza sativa indica DOURADOAGULHA) seedlings were separately 100 ?M arsenite [As(III)] and arsenate [As(V)] in hydroponic culture for 12 h. Genome-wide transcriptomic analysis revealed that 2983 1844 genes responsive under As(III) As(V) stresses, respectively. There 915 shared between treatments. Arsenic stress induced changes complicated gene regulatory pathway seedlings, including transcriptional regulation, hormone signaling, redox, transporters detoxification process. In 12, 24, 48 72 h, qRT-PCR showed WRKY4, bHLH AP2-EREBP associated with regulation significantly down-regulated, while NAC [NAM (no apical meristem), ATAF (Arabidopsis thaliana activating factor) CUC (cup-shaped cotyledon)] transcription factor was up-regulated. The signaling related OZG2, redox encoding Prx, GST, oxidoreductase cytochrome P450, transporter process OsABCC9 ZIP3 affected As stress. Under treatments 1 3 d, malondialdehyde (MDA) contents increased, catalase (CAT), peroxidase (POD), superoxide dismutase (SOD) glutathione reductase (GR) declined. addition, SOD, Grx TrxR mostly down-regulated. conclusion, combination transcriptome physiological may provide a deeper understanding about mechanism rice.
